By L. F. Gram, L. P. Balant, Herbert Y. Meltzer, S. G. Dahl

This e-book comprises the papers from invited academics in addition to chosen contributions offered on the sixth overseas assembly on medical Pharmacology in Psychiatry (I.M.C.P.P.) held in Geneva, Switzerland, 5-7 June 1991. At this assembly the fundamental subject matter of the former conferences during this sequence (Chicago 1979, Troms0 1980, Odense 1982, Bethesda 1985, Troms0 1988) was once persisted, specifically, to bridge the space among experimental improvement and scientific fact in psychopharmacology. After greater than 25 years of extensive study in organic psychiatry, easy realizing of the organic mechanisms underlying significant psychiatric ailments has complex considerably yet remains to be faraway from whole. Likewise, the hypotheses underlying the advance of recent psychotropics were subtle and produced a large spectrum of novel, but designed compounds. The the most important situation for all growth during this box is trustworthy, informative medical trying out of latest compounds. it's our desire that this booklet, as a continuation of the sooner guides during this sequence, presents additional proof of the continued interplay among preclinical and scientific scientists, who merely jointly can guarantee development during this intriguing sector of analysis and medical practice.

ROTH l 1 Introduction Serotonin signals in neurons are transduced by a large and rapidly growing family of receptors. To date, four principal types of serotonin receptor have been identified, designated 5-HTl -5-HT4. 5-HTl receptors have high (nanomolar) affinity for serotonin. Three 5-HTl subtypes have been identified by molecular cloning: 5-HTla (Fargin et al. 1988; Albert et al. 1990), 5-HTlc (Julius et al. 1988), and 5HTld . In addition, 5-HTlb and 5-HTle receptors have been characterized pharmacologically, but their sequences have not yet been deduced from cloned cDNAs.

Evidence for the heterogeneity of serotonin (5-hydroxytryptamine; 5-HT) receptors and recent progress in their classification (reviewed by Bradley et al. 1986; Peroutka 1988; Frazer et al. 1990; Gathert 1990a) have promoted the development of selective agonists and antagonists directed at certain 5-HT receptor classes and subclasses. Some of these drugs are considered as potentially useful for the treatment of psychiatric disorders, for example, 5-HT1A receptor agonists, which act as anxiolytics and antidepressants, and 5-HT3 receptor antagonists, which appear to possess antipsychotic properties.
